Pondexter was selected by the Oklahoma City Thunder with the 26th overall pick in the 2010 NBA draft. After a month, the New Orleans Hornets acquired Pondexter and Craig Brackins.

Prior to the start of the 2011-12 NBA season, the Hornets traded him to the Memphis Grizzlies. After two seasons with the team, he signed a four-year, $14 million deal with the Grizzlies in 2013. Pondexter appeared in 189 games during his first three NBA seasons. He did, however, suffer an injury to his right leg during the 2013-14 NBA season, and he ended up missing the remainder of the season following December surgery. Pondexter returned to the Grizzlies for the 2014-15 season, appearing in the first two months of the season.

He was then traded back to New Orleans, now known as the Pelicans, in a three-team deal involving the Grizzlies and the Boston Celtics. He scored an average of 9 points per game and scored a career-high 25 points against the Brooklyn Nets.

He was forced to retire due to a series of injuries but returned following a series of surgeries. Similarly, he was acquired by the Chicago Bulls in 2017 and faced the Toronto Raptors while nursing a left hamstring injury. He was waived by the Bulls.
